Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003036.4(SKI):c.2049_2090del (p.Gln683_Glu696del), citing Ambry Variant Classification Scheme 2023: The c.2049_2090del42 variant (also known as p.Q683_E696del) is located in coding exon 7 of the SKI gene. This variant results from an in-frame GCTGCGGGCCGACCTGCTGCGGGAGCGCGAGGCCCGGGAGCA deletion at nucleotide positions 2049 to 2090. This results in the in-frame deletion of 14 amino acids from codon 683 to codon 696. This amino acid region is well conserved in available vertebrate species. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
