Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.1075T>C (p.Cys359Arg), citing Ambry Variant Classification Scheme 2023: The p.C359R variant (also known as c.1075T>C), located in coding exon 7 of the DICER1 gene, results from a T to C substitution at nucleotide position 1075. The cysteine at codon 359 is replaced by arginine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
