Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.2002G>C (p.Asp668His), citing Ambry Variant Classification Scheme 2023: The p.D668H variant (also known as c.2002G>C) is located in coding exon 18 of the NF1 gene. The aspartic acid at codon 668 is replaced by histidine, an amino acid with similar properties. This change occurs in the first base pair of coding exon 18. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
