Uncertain significance — the classification assigned by Ambry Genetics to NM_002519.3(NPAT):c.2002G>A (p.Glu668Lys), citing Ambry Variant Classification Scheme 2023: The p.E668K variant (also known as c.2002G>A), located in coding exon 13 of the NPAT gene, results from a G to A substitution at nucleotide position 2002. The glutamic acid at codon 668 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
