NM_000535.7(PMS2):c.1997A>G (p.Lys666Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K666R variant (also known as c.1997A>G), located in coding exon 11 of the PMS2 gene, results from an A to G substitution at nucleotide position 1997. The lysine at codon 666 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
