NM_001267550.2(TTN):c.47060G>A (p.Ser15687Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S6622N variant (also known as c.19865G>A), located in coding exon 79 of the TTN gene, results from a G to A substitution at nucleotide position 19865. The serine at codon 6622 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
