Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001244008.2(KIF1A):c.1984C>T (p.Arg662Trp), citing Ambry Variant Classification Scheme 2023: The p.R662W variant (also known as c.1984C>T), located in coding exon 21 of the KIF1A gene, results from a C to T substitution at nucleotide position 1984. The arginine at codon 662 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
