Uncertain significance for Familial adenomatous polyposis 4 — the classification assigned by St. Jude Molecular Pathology, St. Jude Children's Research Hospital to NM_002439.5(MSH3):c.197C>T (p.Pro66Leu), citing St. Jude Assertion Criteria 2020: The MSH3 c.197C>T (p.Pro66Leu) missense change is absent in gnomAD v2.1.1 (https://gnomad.broadinstitute.org/), however data at this position may not be reliable due to mean depth of coverage <30X. The in-silico tool REVEL predicts a benign effect on protein function, but this prediction has not been confirmed by functional studies. This variant has not been reported in individuals with MSH3-associated polyposis syndrome. In summary, the evidence currently available is insufficient to determine the clinical significance of this variant. It has therefore been classified as of uncertain significance.
