Uncertain significance — the classification assigned by Ambry Genetics to NM_002519.3(NPAT):c.197A>G (p.Tyr66Cys), citing Ambry Variant Classification Scheme 2023: The p.Y66C variant (also known as c.197A>G), located in coding exon 3 of the NPAT gene, results from an A to G substitution at nucleotide position 197. The tyrosine at codon 66 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
