NM_006073.4(TRDN):c.1975+1G>C was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the TRDN gene (transcript NM_006073.4) at the canonical splice donor site of the intron immediately after coding-DNA position 1975, where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.1975+1G>C intronic variant results from a G to C substitution one nucleotide after coding exon 38 of the TRDN gene. This nucleotide position is well conserved in available vertebrate species. Using the BDGP and ESEfinder splice site prediction tools, this alteration is predicted to abolish the native splice donor site; however, direct evidence is unavailable. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. However, this alteration does not impact the predominant cardiac isoform of TRDN (NM_001256021.1; Kobayashi YM et al. J. Biol. Chem., 1999 Oct;274:28660-8). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
