Uncertain significance — the classification assigned by Ambry Genetics to NM_021930.6(RINT1):c.1974T>G (p.His658Gln), citing Ambry Variant Classification Scheme 2023: The p.H658Q variant (also known as c.1974T>G), located in coding exon 13 of the RINT1 gene, results from a T to G substitution at nucleotide position 1974. The histidine at codon 658 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
