NM_198578.4(LRRK2):c.1970A>T (p.Lys657Ile) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K657I variant (also known as c.1970A>T), located in coding exon 17 of the LRRK2 gene, results from an A to T substitution at nucleotide position 1970. The lysine at codon 657 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
