Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.1964G>A (p.Gly655Glu), citing Ambry Variant Classification Scheme 2023: The p.G655E variant (also known as c.1964G>A), located in coding exon 17 of the NF1 gene, results from a G to A substitution at nucleotide position 1964. The glycine at codon 655 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
