NM_004656.4(BAP1):c.1959G>C (p.Glu653Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E653D variant (also known as c.1959G>C), located in coding exon 15 of the BAP1 gene, results from a G to C substitution at nucleotide position 1959. The glutamic acid at codon 653 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
