This variant lies in the MSH2 gene (transcript NM_000251.3) at coding-DNA position 1957, where A is replaced by C; at the protein level this means replaces asparagine at residue 653 with histidine — a missense variant. Submitter rationale: The p.N653H variant (also known as c.1957A>C), located in coding exon 12 of the MSH2 gene, results from an A to C substitution at nucleotide position 1957. The asparagine at codon 653 is replaced by histidine, an amino acid with similar properties. In a massively parallel cell-based functional assay testing susceptibility to a DNA damaging agent, 6-thioguanine (6-TG), this variant was determined to be functionally neutral (Jia X et al. Am J Hum Genet, 2021 01;108:163-175). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
