Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.2090A>G (p.Asp697Gly), citing Ambry Variant Classification Scheme 2023: The p.D651G variant (also known as c.1952A>G), located in coding exon 19 of the KIF1B gene, results from an A to G substitution at nucleotide position 1952. The aspartic acid at codon 651 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
