Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_017636.4(TRPM4):c.1946G>T (p.Trp649Leu), citing Ambry Variant Classification Scheme 2023: The p.W649L variant (also known as c.1946G>T), located in coding exon 14 of the TRPM4 gene, results from a G to T substitution at nucleotide position 1946. The tryptophan at codon 649 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
