The p.Val623Ile var iant in CDH23 has been previously reported by our laboratory in the homozygous s tate in 1 individual with hearing loss with delayed walking; however, this patie nt harbored a homozygous likely pathogenic variant in another gene that explaine d the phenotype. This variant has also been identified in 0.13% (39/29954) of So uth Asian chromosomes by the Genome Aggregation Database (gnomAD, http://gnomad. broadinstitute.org/; dbSNP rs143782870); however its frequency is not high enoug h to rule out a pathogenic role. Computational prediction tools and conservation analysis do not provide strong support for or against an impact to the protein.
