Uncertain significance — the classification assigned by Ambry Genetics to NM_003579.4(RAD54L):c.1934T>G (p.Val645Gly), citing Ambry Variant Classification Scheme 2023: The p.V645G variant (also known as c.1934T>G), located in coding exon 17 of the RAD54L gene, results from a T to G substitution at nucleotide position 1934. The valine at codon 645 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
