NM_000400.4(ERCC2):c.1931T>A (p.Phe644Tyr) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F644Y variant (also known as c.1931T>A), located in coding exon 21 of the ERCC2 gene, results from a T to A substitution at nucleotide position 1931. The phenylalanine at codon 644 is replaced by tyrosine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
