Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.1930A>G (p.Asn644Asp), citing Ambry Variant Classification Scheme 2023: The p.N644D variant (also known as c.1930A>G), located in coding exon 17 of the BUB1 gene, results from an A to G substitution at nucleotide position 1930. The asparagine at codon 644 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
