Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.21533-2384G>T, citing Ambry Variant Classification Scheme 2023: The p.D6434Y variant (also known as c.19300G>T), located in coding exon 81 of the OBSCN gene, results from a G to T substitution at nucleotide position 19300. The aspartic acid at codon 6434 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
