Uncertain significance — the classification assigned by Ambry Genetics to NM_001386125.1(OBSCN):c.21533-2456T>C, citing Ambry Variant Classification Scheme 2023: The p.W6410R variant (also known as c.19228T>C), located in coding exon 81 of the OBSCN gene, results from a T to C substitution at nucleotide position 19228. The tryptophan at codon 6410 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
