NM_004830.4(MED23):c.191T>C (p.Val64Ala) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V64A variant (also known as c.191T>C), located in coding exon 4 of the MED23 gene, results from a T to C substitution at nucleotide position 191. The valine at codon 64 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
