Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.1906del (p.His636fs), citing Ambry Variant Classification Scheme 2023: The c.1906delC pathogenic mutation, located in coding exon 12 of the ATM gene, results from a deletion of one nucleotide at nucleotide position 1906, causing a translational frameshift with a predicted alternate stop codon (p.H636Tfs*13). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
