Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.1903G>T (p.Gly635Cys), citing Ambry Variant Classification Scheme 2023: The p.G635C variant (also known as c.1903G>T), located in coding exon 14 of the RECQL gene, results from a G to T substitution at nucleotide position 1903. The glycine at codon 635 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
