Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.1897T>C (p.Cys633Arg), citing Ambry Variant Classification Scheme 2023: The p.C633R variant (also known as c.1897T>C), located in coding exon 17 of the BUB1 gene, results from a T to C substitution at nucleotide position 1897. The cysteine at codon 633 is replaced by arginine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
