Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000179.3(MSH6):c.1892C>G (p.Ser631Cys), citing Ambry Variant Classification Scheme 2023: The p.S631C variant (also known as c.1892C>G), located in coding exon 4 of the MSH6 gene, results from a C to G substitution at nucleotide position 1892. The serine at codon 631 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
