NM_003239.5(TGFB3):c.188C>T (p.Thr63Ile) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T63I variant (also known as c.188C>T), located in coding exon 1 of the TGFB3 gene, results from a C to T substitution at nucleotide position 188. The threonine at codon 63 is replaced by isoleucine, an amino acid with similar properties. A different variant affecting this codon (p.T63N, c.188C>A) has been detected in an arrhythmogenic right ventricular cardiomyopathy cohort; however, details were limited (Bao J et al. Circ Cardiovasc Genet, 2013 Dec;6:552-6). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
