Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.1880T>A (p.Ile627Asn), citing Ambry Variant Classification Scheme 2023: The p.I627N variant (also known as c.1880T>A), located in coding exon 10 of the DICER1 gene, results from a T to A substitution at nucleotide position 1880. The isoleucine at codon 627 is replaced by asparagine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
