Submitter rationale: The c.1880_1881insGGA variant (also known as p.F626_D627insE), located in coding exon 14 of the VCL gene, results from an in-frame GGA insertion at nucleotide positions 1880 to 1881. This results in the insertion of an extra glutamic acid residue between codons 626 and 627. This amino acid region is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
