Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_017636.4(TRPM4):c.1875C>G (p.Asp625Glu), citing Ambry Variant Classification Scheme 2023: The p.D625E variant (also known as c.1875C>G), located in coding exon 14 of the TRPM4 gene, results from a C to G substitution at nucleotide position 1875. The aspartic acid at codon 625 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
