Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004064.5(CDKN1B):c.186C>A (p.Phe62Leu), citing Ambry Variant Classification Scheme 2023: The p.F62L variant (also known as c.186C>A), located in coding exon 1 of the CDKN1B gene, results from a C to A substitution at nucleotide position 186. The phenylalanine at codon 62 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
