NM_004655.4(AXIN2):c.1867A>C (p.Met623Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M623L variant (also known as c.1867A>C), located in coding exon 6 of the AXIN2 gene, results from an A to C substitution at nucleotide position 1867. The methionine at codon 623 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
