Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003036.4(SKI):c.1861G>A (p.Ala621Thr), citing Ambry Variant Classification Scheme 2023: The p.A621T variant (also known as c.1861G>A), located in coding exon 6 of the SKI gene, results from a G to A substitution at nucleotide position 1861. The alanine at codon 621 is replaced by threonine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
