Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002485.5(NBN):c.1859T>C (p.Ile620Thr), citing Ambry Variant Classification Scheme 2023: The p.I620T variant (also known as c.1859T>C), located in coding exon 12 of the NBN gene, results from a T to C substitution at nucleotide position 1859. The isoleucine at codon 620 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
