NM_007294.4(BRCA1):c.1859T>C (p.Ile620Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I620T variant (also known as c.1859T>C), located in coding exon 9 of the BRCA1 gene, results from a T to C substitution at nucleotide position 1859. The isoleucine at codon 620 is replaced by threonine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
