NM_000384.3(APOB):c.1858A>G (p.Lys620Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K620E variant (also known as c.1858A>G), located in coding exon 14 of the APOB gene, results from an A to G substitution at nucleotide position 1858. The lysine at codon 620 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
