NM_001365951.3(KIF1B):c.1974C>G (p.Asp658Glu) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D612E variant (also known as c.1836C>G), located in coding exon 18 of the KIF1B gene, results from a C to G substitution at nucleotide position 1836. The aspartic acid at codon 612 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
