NM_181486.4(TBX5):c.1063C>T (p.Arg355Cys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R355C variant (also known as c.1063C>T), located in coding exon 8 of the TBX5 gene, results from a C to T substitution at nucleotide position 1063. The arginine at codon 355 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant was reported in an early onset atrial fibrillation cohort, and overexpression studies suggested possible functional impact (Ma JF et al. Cardiovasc Res, 2016 Mar;109:442-50). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
