Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_002691.4(POLD1):c.1831A>G (p.Met611Val), citing Ambry Variant Classification Scheme 2023: The p.M611V variant (also known as c.1831A>G), located in coding exon 14 of the POLD1 gene, results from an A to G substitution at nucleotide position 1831. The methionine at codon 611 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
