This variant lies in the DDC gene (transcript NM_001082971.2) at coding-DNA position 304, where G is replaced by A; at the protein level this means replaces glycine at residue 102 with serine — a missense variant. Submitter rationale: This missense change has been observed in individuals with aromatic L-amino acid decarboxylase deficiency (PMID: 9789536, 14991824, 28856607, 32409695). It has also been observed to segregate with disease in related individuals. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on DDC protein function. Experimental studies have shown that this missense change affects DDC function (PMID: 21541720, 24865461). For these reasons, this variant has been classified as Pathogenic. This variant is present in population databases (rs137853207, gnomAD 0.003%). This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 102 of the DDC protein (p.Gly102Ser).
