Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_012144.4(DNAI1):c.1823A>C (p.His608Pro), citing Ambry Variant Classification Scheme 2023: The p.H608P variant (also known as c.1823A>C), located in coding exon 19 of the DNAI1 gene, results from an A to C substitution at nucleotide position 1823. The histidine at codon 608 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
