Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_024301.5(FKRP):c.180C>G (p.Asp60Glu), citing Ambry Variant Classification Scheme 2023: The p.D60E variant (also known as c.180C>G), located in coding exon 1 of the FKRP gene, results from a C to G substitution at nucleotide position 180. The aspartic acid at codon 60 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
