NM_001365951.3(KIF1B):c.1946C>G (p.Ser649Cys) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S603C variant (also known as c.1808C>G), located in coding exon 18 of the KIF1B gene, results from a C to G substitution at nucleotide position 1808. The serine at codon 603 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
