This variant lies in the SMARCA4 gene (transcript NM_003072.5) at coding-DNA position 1795, where A is replaced by G; at the protein level this means replaces isoleucine at residue 599 with valine — a missense variant. Submitter rationale: The p.I599V variant (also known as c.1795A>G), located in coding exon 10 of the SMARCA4 gene, results from an A to G substitution at nucleotide position 1795. The isoleucine at codon 599 is replaced by valine, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be benign and tolerated by PolyPhen and SIFT in silico analyses, respectively. Since supporting evidence is limited at this time, the clinical significance of p.I599V remains unclear.
