NM_000251.3(MSH2):c.1059G>T (p.Lys353Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K353N variant (also known as c.1059G>T), located in coding exon 6 of the MSH2 gene, results from a G to T substitution at nucleotide position 1059. The lysine at codon 353 is replaced by asparagine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
