NM_001927.4(DES):c.172C>T (p.Arg58Cys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R58C variant (also known as c.172C>T), located in coding exon 1 of the DES gene, results from a C to T substitution at nucleotide position 172. The arginine at codon 58 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species; however, cysteine is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
