Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_003042.4(SLC6A1):c.1721G>C (p.Ser574Thr), citing Ambry Variant Classification Scheme 2023: The p.S574T variant (also known as c.1721G>C), located in coding exon 14 of the SLC6A1 gene, results from a G to C substitution at nucleotide position 1721. The serine at codon 574 is replaced by threonine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
