NM_017950.4(CCDC40):c.1715G>C (p.Cys572Ser) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C572S variant (also known as c.1715G>C), located in coding exon 11 of the CCDC40 gene, results from a G to C substitution at nucleotide position 1715. The cysteine at codon 572 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
